Extraction of Polysaccharides from Endoconcha Sepiae by Hot Water Extraction

Objective: to extract polysaccharide active substance from Endoconcha Sepiae (Endoconcha Sepiae) and study its properties and medicinal value. Methods: crude Endoconcha Sepiae polysaccharide was extracted by hot water extraction, CPS-1 and CBP-S were obtained after purification, and the content, purity and relative molecular weight of the developed polysaccharide were determined. The extracted Endoconcha Sepiae polysaccharide was used to establish mouse UC type by DSS induction method. SMZ-TMP was used as control. The protective effect of high and low dose CPS-1 on gastric ulcer in UC mice was studied by clinical symptoms, body mass change, determination of EGF, TNG-α, PDGF content in blood, acute toxicity study, pH value of gastric content and other indicators. Results: 85℃ was the best extraction temperature; the best extraction condition of polysaccharide was smashing for 5 min, adding 200 mL distilled water per 50 g Endoconcha Sepiae powder, extracting for 6 h at 85℃, the extraction yield of polysaccharide was 2‰. After further purification and activity tracking on DEAE Sepharose fast flow, Sepharose CL-6B column and Sephacr-yl S-300 column, a relatively uniform active fine product was obtained. The relative molecular weight of CPS-1 and CPW-2 is 1×106 and 1.1 × 105, respectively. Endoconcha Sepiae polysaccharide CPS-1 can reduce the effect of UC on the body weight of experimental mice, and significantly improve the expression of EGF and PDGF in the blood of UC mice. The administration time increased from 2 days to 4 days, and the same concentration of 400 mg/kg CBP-S had no significant difference in improving the pH value of gastric acid in mice. Conclusion: different extraction conditions affect the yield of Endoconcha Sepiae polysaccharide. The separation materials with different properties can separate the polysaccharides with different molecular weight and charge, and achieve the purpose of separation. CPS-1, a high-quality polysaccharide, is a homogenous polysaccharide component obtained from Endoconcha Sepiae, a marine Chinese herbal medicine. Endoconcha Sepiae polysaccharide CPS-1 and CBP-S can accelerate the healing and repair process of ulcer tissue and play a protective role on gastric mucosa of mice.
